Hantaviruses, classified as Category A pathogens by the U.S. Centers for Disease Control and Prevention (CDC), are emerging zoonotic pathogens belonging to the Bunyaviridae family. The tripartite negative-strand RNA genome of hantaviruses consists of three genomic RNA segments (S, M, and L) that encode the nucleocapsid protein (N), a precursor glycoprotein (which forms envelope glycoproteins Gn and Gc), and the viral RNA-dependent RNA polymerase (RdRp). The N protein serves as the primary structural component, protecting and encapsulating the genomic RNA strands to form viral ribonucleocapsids. Recent studies indicate that the N protein acts in concert with RdRp to play a crucial role in viral transcription and replication, and it preferentially promotes the translation of viral mRNA within the cell.

Currently, no antiviral therapies or vaccines are available for hantavirus-associated diseases. A deeper understanding of the molecular mechanisms underlying the hantavirus life cycle could help identify targets for antiviral therapy and design potential drugs to treat HFRS and HCPS. Given the alarming case-fatality rates, the development of an effective hantavirus vaccine is imperative. Additionally, because early clinical manifestations of hantavirus infection often resemble common seasonal respiratory or influenza-like illnesses, there is an urgent need for highly sensitive and reliable laboratory diagnostic tools to enable early differential diagnosis and accurate target validation.
